A complete microprocessor controlled Uv-irradiation system, mainly dedicated to the linking of nucleic acid to membranes and elimination of PCR contamination.
Application
    1 - Crosslinking of DNA or RNA by covalently binding nucleic acids to nitrocellulose or nylon membranes for southern,  Northern, dot or slot blot procedures.
    2 - Partial restriction endonuclease digestion by formation of thymine dimmers.
    3 - Rec A mutation screening.

The polymerase chain reaction (PCR) is a biochemical technology used  in molecular biology to amplify a single or few copies of a piece of DNA  across several orders of magnitude , generating thousands to millions of copies of a particular DNA sequence . The method relies on thermal cycling consisting of cycles of repeated heating and cooling of the reaction for DNA melting and enzymatic replication of DNA. Primers (short DNA fragments) containing sequences complementary to the target region along with DNA Polymerase ) are key components to enable
the
 amplification .

Electrophoresis is a technique used to separate and test purity of macromolecules - especially proteins and nucleic acids - that differ in size, charge or conformation. When charged molecules are placed in an electric field, they migrate towards either the positive or negative pole according to their charge. Proteins and nucleic acids are electrophoresed within a matrix or "gel" This gel is composed of either agarose for nucleic acids or polyacrylamide for protein.
The gel is immersed within an electrophoresis buffer that provides ions to carry a current and some type of buffer to maintain the pH at a relatively constant value.

Gel Documentation system

A gel doc is an equipment widely used for imaging and documentation of nucleic acid and protein suspended within agarose or polyacrylamide gels. These gels are stained with ethidium bromide or other phlorophores such as SYBR Green. The fluorescent substance that has bounded to nucleic acids is excited via ultraviolet irradiation and emits fluorescent light. The amount of the fluorescent substance as ETBr bounded to nucleic acids depend on the molecular weight and concentration of the nucleic acid, in other words, a band of a large molecular weight will shine brighter; conversely fluorescence will be weaker for band of small molecular weight.

Theory:
The NanoDrop Spectrophotometer is used for measuring nucleic acid concentrations in sample volumes of (6) micro liter. This technology overcomes the need for cuvettes when taking measurements. This is accomplished by placing the sample directly on top of the detection surface and using the surface tension to create a column between the ends of optical fibers.
Applications:
- Determination of nucleic acid concentration.
- Dermination of nucleic acid samples purity.
- Purified protein analysis (A280).

REAL- TIME PCR
(Rotor-Gene Q 5plex)

Real-time PCR is technique used to quantify starting amounts of nucleic acid template by analyzing the amount of DNA produced during each cycle of PCR. In this technique the detection of PCR products is made by including in the reaction a fluorescent molecule that reports an increase in the amount of DNA with a proportional increase in fluorescent signal.
Applications:
1. Quantitative mRNA expression studies.
2. DNA copy number measurements in genomic or viral DNAs.
3. Allelic discrimination assays or SNP genotyping.
4. Verification of micro array results.
5. Drug therapy efficacy.
6. DNA damage measurement.
* * In our lab now we are concerned with quantitative mRNA expression studies only.

Comparing the gene expression in different cell lines or tissues. Studying the time course of drug treatment compared to the untreated control and measuring the gene expression in diseased versus non-diseased tissues.

A high-speed centrifuge for subjecting sols or solutions to forces many times that of gravity and producing concentration differences depending on the weight of the micelle or molecule.
Applications:
Ultracentrifuges are available with a wide variety of rotors suitable for a great range of experiments. Preparative rotors are used in biology for pelleting of fine particulate fractions, such as cellular organelles. They can also be used for gradient separations, in which the tubes are filled from top to bottom with an increasing concentration of a dense substance in solution. After the sample has spun at high speed for sufficient time to produce the separation, the rotor is allowed to come to a smooth stop and the gradient is gently pumped out of each tube to isolate the separated components.
